Introducción: En condiciones naturales, las raíces del arbusto, Phyllanthus acuminatus, producen bajas concentraciones de metabolitos secundarios de interés medicinal. Esto abre una oportunidad para el cultivo in vitro, para aumentar la concentración de metabolitos. Objetivo: Determinar las condiciones óptimas de cultivo líquido para raíces pilosas de P. acuminatus. Métodos: Se utilizó la evaluación del crecimiento de la biomasa según porcentaje de inóculo inicial (0.50 y 0.10 %), porcentaje de nutrientes de los medios (100, 50 y 25 %) y tasa de agitación (90, 100 y 110 min-1) (N= 15 repeticiones). Resultados: Las mejores condiciones de cultivo líquido fueron: 0.10 % de inóculo inicial, nutrientes al 25 % y 90 min-1 para la tasa de agitación. Hay diferencias entre las raíces pilosas y las raíces no transformadas. Conclusiones: es factible producir raíces pilosas de P. acuminatus a gran escala, aplicando e implementando las condiciones evaluadas de porcentaje de inóculo, nutrientes en el medio y tasas de agitación utilizadas en este estudio.


Introduction: Under natural conditions, the roots of the shrub, Phyllanthus acuminatus, produce low concentrations of secondary metabolites of medicinal interest. This opens an opportunity for in vitro culture, to increase metabolite concentration. Objective: To determine the optimal liquid culture conditions for hairy roots of P. acuminatus. Methods: We used biomass growth evaluation according to initial inoculum percentage (0.50 and 0.10 %), percentage of medium nutrients (100, 50 and 25 %) and agitation rate (90, 100 and 110 min-1) (N=15 replications). Results: The best liquid culture conditions were: 0.10 % of initial inoculum, nutrients at 25 % and 90 min-1 for the agitation rate. There are differences among hairy roots and non-transformed roots. Conclusions: It is feasible to produce P. acuminatus hairy roots at a large scale, applying and implementing the evaluated conditions of inoculum percentage, nutrients in the medium and agitation rates.

ABSTRACT

Abstract The "arnicas" found in Brazil are examples of different species of the family Asteraceae used in popular medicine for its attributed anti-inflammatory action. Among the species known and used as "arnica" we selected: Calea uniflora Less., Chaptalia nutans (L.) Polák, Lychnophora ericoides Mart., Lychnophora pinaster Mart., Lychnophora salicifolia Mart., Lychnophora diamantinana Coile & S.B.Jones, Porophyllum ruderale (Jacq.) Cass., Pseudobrickellia brasiliensis (Spreng.) R.M.King & H.Rob., Sphagneticola trilobata (L.) Pruski, and Solidago chilensis Meyen, due to their extensive use. This research provides new information on leaf morphology and anatomy and on chemistry of the major metabolites found in these species through histochemical tests and phytochemical review. The results revealed anatomical characters for the differentiation and quality control of the vegetal drugs, being these: distinctive epidermal attachments, epidermis cells, parenchymal cells of the mesophyll, vascular bundles, midvein patterns and secretory structures of exudation of secondary metabolites. The review of chemical profiles showed differences in the chemical composition of the species, as different skeletons of sesquiterpene lactones in the species evaluated in addition to other chemical classes such as terpenes, flavonoids, chromenes and phenolic acids derivate. Based on the results obtained in this work it is important to emphasize that the information about the ten species of arnica generate subsidies for differentiation and identification of characteristic markers and for the diagnosis of the species and it can be applied in the "arnicas" quality control.

ABSTRACT This study aims the anatomical description and chemical characterization of aerial parts of Clusia criuva Cambess., Clusiaceae in addition to the evaluation of the antioxidant activity of crude extracts, correlated to the flavonoid content. The morphological characterization was performed using traditional techniques of plant anatomy. For phytochemical studies, crude extracts were obtained by static maceration and analyzed by thin layer chromatography. The antioxidant activity and the flavonoids content were determined by colorimetric methods involving, respectively, 2,2-diphenyl-1-picrylhydrazyl free radical and aluminum chloride. C. criuva has uniseriate epidermis, paracytic stomata, hypostomatic leaves, cuticular flanges and cordiform vascular cylinder with accessory bundles. Chemical prospecting confirmed the abundant presence of terpenes and phenols in the extracts of leaves and of fruits. The methanolic extract of seeds showed the lowest EC50 value, but the methanolic extract of pericarps exhibited the highest maximum antioxidant activity. The results suggested a high percentage of flavonoids in the hexanic extract of pericarps, however, this could represent, in fact, the presence of benzophenones. Secretory ducts and the shape of the midrib are diagnostic for C. criuva. The antioxidant activity is not directly related to the flavonoids. The results indicate the importance of future studies with C. criuva chemical constituents.

ABSTRACT

ABSTRACT Baccharis belongs to the Asteraceae family and comprises a number of medicinal species. Baccharis brevifolia DC., B. microdonta DC., B. pauciflosculosa DC., and B. trilobata A.S. Oliveira & Marchiori, which are popularly known in Brazil as “vassouras” (“broom”), are all found in Southern Brazil. The anatomical features of the leaf and stem were investigated by employing the usual light and scanning electron microtechniques, as a means of differentiating the taxa. The following anatomical characteristics can be considered to be diagnostic: the occurrence and type of stomata, midrib, stem and crystal shapes, and the presence of the petiole.

ABSTRACT Cissampelos pareira L., belonging to Menispermaceae family, has worldwide distribution, occurring in tropical and subtropical regions of the Americas, Africa and Asia. It is the most popular species of Cissampelos, known for its medicinal uses of leaves and roots. The study aims to find distinctive leaf anatomical characters, and also demonstrate the importance of spectral data to identify C. pareira samples, in order to contribute to its taxonomy and quality control of its drugs. Anatomical leaf analyses were performed by optical and scanning electron microscopy. The spectral profile was obtained from methanolic extracts of C. pareira samples from Brazil and Africa, with application of UV–vis spectrophotometry data, which were analyzed by principal component analysis (PCA). Some anatomical characters such as leaf epidermal cells walls, stomata, trichomes, mesophyll, features of midrib and petiole, and the spectral profile within the wavelength ranging between 770 and 240 nm (eight bands) differs between Brazilian and African samples. The results represent an additional support to the taxonomy of C. pareira, and the quality control of their leaf drugs, mainly in relation to misidentified samples.

Abstract Amphilophium crucigerum (L.) L.G. Lohmann, known as “pente-de-macaco” is a species of Bignoniaceae native to Brazil, and whose seeds are used in folk medicine. This study aimed to describe morphoanatomical features of this species of fruit to aid in its correct identification and pharmacognostic analysis. Samples of ovary, pericarp and seed were fixed with 3% glutaraldehyde, sectioned on a rotary microtome and analyzed by stereomicroscope. The results are shown in three parts: (1) The ovary presents peltate trichomes, long non-glandular trichomes and emergences in the epidermis; it is 2-carpellate and unilocular with two intruding parietal placenta; ovules are numerous on the placenta; it presents a large quantity of crystals. (2) The pericarp is woody, densely echinate and elliptic shape; it presents a 2-valved capsule and is septicidal; it presents emergences, stomata, lenticels, crystals and a large quantity of clustered stones cells. (3) Seeds are alate, exalbuminate and exotestal; there is a large amount of crystals in the exotestal region; it presents an endothelium and remnant endosperm. Histochemical tests showed the presence of lipophilic substances, polysaccharides, phenolic substances, alkaloids and a small quantity of starch. These pharmacobotanical features described for A. crucigerum are essential for the pharmacognostic analysis of the drug plant.

RESUMO Tanacetum vulgare L., conhecida popularmente como catinga-de-mulata, é utilizada na medicina tradicional como vermífugo, digestivo e emenagogo. Objetivou-se analisar a morfoanatomia de folhas e caules dessa espécie medicinal com vistas ao controle da qualidade. Foram utilizadas técnicas usuais de microscopia de luz e eletrônica de varredura. As folhas são compostas, pinatissectas e anfiestomáticas. Tricomas tectores flageliformes simples e glandulares capitados são observados. A nervura central tem formato biconvexo, o pecíolo é côncavo-convexo, a ráque é plano-convexa e o caule é arredondado. Feixes vasculares colaterais, calotas de fibras perivasculares e colênquima lamelar estão presentes na folha e no caule. Os caracteres morfoanatômicos evidenciados contribuem na identificação do táxon e fornecem subsídios farmacobotânicos para o controle da qualidade de drogas vegetais e fitoterápicos.


ABSTRACT Tanacetum vulgare L., popularly known as tansy, is used in traditional medicine as a vermifuge, digestive and emmenagogue. This study aimed to analyze the morphoanatomical leaves and stems of this medicinal plant in order to control the quality. Usual techniques of light and scanning electron microscopy were used. The leaves are composed, pinatissect and amphistomatic. Simple and flagelliform non- glandular trichomes and capitate glandular trichomes are observed. The midrib has biconvex shape, the petiole is concavo-convex, the rachis is plano-convex, and the stem is rounded. Collateral vascular bundles, perivascular fiber caps and laminar collenchyma are encountered in the leaves and stems. Evidenced the morphological and anatomical features contribute to the identification of the taxon and provide pharmacobotanical data for the quality control of herbal drugs.

AbstractBaccharis L. sect. Caulopterae, Asteraceae, comprises thirty species in Brazil that show stems represented by cladodes, which are very similar in morphology. These species are popularly known as “carqueja” in Brazil and Argentina and are used in popular medicine as diuretic and stomachic. The aim of this work was to examine the morpho-anatomical characters of cladodes of Baccharis pentaptera (Less.) DC. for diagnosis purposes. The plant material was prepared by light and scanning electron microscopy. B. pentaptera shows opposite and spread wings in the two-winged cladode axis and irregular arrangement in the three-winged cladode. The wings have a uniseriate epidermis with palisade parenchyma next to both sides of epidermis. The spongy parenchyma crossed by minor collateral vascular bundles is observed in the central region of wings. The glandular trichomes are capitate and biseriate and the non-glandular trichomes are uniseriate and flagelliform with 2–3 cells that extend from the base. In caulinar axis, there are uniseriate epidermis, chlorenchyma alternating with angular collenchyma and perivascular fiber caps adjoining the phloem which is outside the xylem. Prismatic and styloid crystals are verified in the perimedullary zone. These combined characters can assist the diagnosis of Baccharis species sect. Caulopterae.

AbstractDetermining the authenticity and quality of plant raw materials used in the formulation of herbal medicines, teas and cosmetics is essential to ensure their safety and efficacy for clinical use. Some Passiflora species are officially recognized in the pharmaceutical compendia of various countries and have therapeutic uses, particularly as sedatives and anxiolytics. However, the large number of Passiflora species, coupled with the fact that most species are popularly known as passion fruit, increases the misidentification problem. The purpose of this study is to make a pharmacognostic comparison between various Passiflora species to establish a morpho-anatomical profile that could contribute to the quality control of herbal drug products that contain passion fruit. This was conducted by collecting samples of leaves from twelve Passiflora taxa (ten species and two forms of P. edulis): P. actinia, P. alata, P. amethystina, P. capsularis, P. cincinnata, P. edulisf. flavicarpa, P. edulis f. edulis, P. incarnata, P. morifolia, P. urnifolia, P. coccinea and P. setacea, from different locations and their morpho-anatomical features were analyzed using optical microscopy and scanning electron microscopy. Microscopic analysis allowed to indicate a set of characters that can help to differentiate species. These include midrib and petiole shape, midrib and petiole vascular pattern, medium vein shape, presence of trichomes, presence of blade epidermal papillae and sclerenchymatic cells adjoining the vascular bundles. These characters could be used to assist in the determination of herbal drug quality and authenticity derived from a species of Passiflora.

Schinus terebinthifolius Raddi (Anacardiaceae), conhecida como aroeira-vermelha é uma espécie nativa do Brasil, perenifólia e pioneira, sendo muito utilizada para combater diversas doenças, pois apresenta, entre outras, propriedades anti-inflamatórias e cicatrizantes. O objetivo desta pesquisa foi realizar um estudo morfoanatômico do caule e da folha de S. terebinthifolius, descrevendo a organização dos tecidos e suas possíveis variações. As análises macro e microscópicas foram realizadas com material selecionado de partes vegetativas de um espécime localizado na cidade de Campina Grande - PB. A aroeira-vermelha é um vegetal arbóreo e dióico, que apresenta caule cilíndrico e simpodial, com casca rugosa e estriada. A folha é perene, alterna, coriácea e composta. As análises microscópicas evidenciaram epiderme caulinar e foliar uniestratificadas, exceto na face adaxial da folha, que é bisseriada. No caule e no pecíolo são visualizados muitos tricomas tectores unicelulares, curtos, cônicos e agudos no ápice. Encontram-se canais secretores arredondados e bem desenvolvidos na região do floema em todos os órgãos estudados, bem como cutícula revestindo as células epidérmicas e grande quantidade de drusas de oxalato de cálcio. Os feixes vasculares são circundados por células esclerenquimáticas e apresentam o floema seguido por xilema helicoidal bem desenvolvido. No caule, pecíolo e nervura central da folha, o xilema é interno e o floema, externo. A aroeira-vermelha apresenta alguns aspectos estruturais característicos de plantas xerófilas, como camada múltipla da epiderme da face adaxial foliar, que protegem o mesofilo de excessiva dessecação; presença de cutícula espessa e grande quantidade de pêlos e de estômatos; além de sistema vascular bem desenvolvido, com abundância de esclerênquima, dando sustentação ao feixe.


Schinus terebinthifolius Raddi (Anacardiaceae), popularly known as Brazilian pepper, is an evergreen, pioneer and indigenous plant from Brazil, and it is used to combat various diseases, as it presents anti-inflammatory and wound healing properties. The aim of this research was to study the morpho-anatomical features of the stem and leaf of S. terebinthifolius, describing the organization of the tissues and their variations. The macroscopic and microscopic analyses were performed with material selected from vegetative parts of a specimen collected in the city of Campina Grande - state of Paraíba, Brazil. The Brazilian pepper is a dioecious plant with cylindrical and sympodial stem and rough and striated bark. The leaves are evergreen, alternate, leathery and compound. The microscopic analysis showed uniseriate epidermis in the stem and leaf, except in the adaxial leaf, which is biseriate. In the stem and petiole, many unicellular, short, conical tector trichomes with acute apex are observed. In the phloem of all studied organs, we found rounded and well-developed secretory ducts, as well as cuticle covering the epidermal cells and a large amount of calcium oxalate druses. The vascular bundles are surrounded by sclerenchyma cells and present phloem followed by well-developed spiral xylem. In the stem, petiole and leaf's central vein, the xylem is internal and the phloem is external. The Brazilian pepper has some structural features characteristic of xerophilous plants, such as multiple layers of the epidermis in adaxial leaf, which protects the mesophyll from excessive drying, thick cuticle and a large quantity of hairs and stomata, well-developed vascular system with plenty of sclerenchyma, giving support to the bundle.

ABSTRACT Tabernaemontana catharinensis A. DC (Apocynaceae) is used as a medicinal plant by the population. In order to contribute to the safe use of the plant as herbal medicine, this study aimed to morphoanatomically characterize the aereal vegetative organs of T. catharinensis and to evaluate the leaves’ mutagenic and antimutagenic activities. Histological blades of leaves and stem of T. catharinensis were performed; the methionine system (methG1) and Aspergillusnidulans conidia germination analysis were employed for mutagenic and antimutagenic evaluation. The morphoanatomic analysis did not show trichomes in the stem, petiole and leaf. Besides, it was observed both the presence of bi-collateral bundles - except in the foliar apex where the bundles were from the collateral type - as well as anamphistomatic leaf with paracyte stomata and sub-epidermal layer in the region of the leaf edges. The mutagenicity/antimutagenicity trial indicated a significant decrease of mutation frequency in comparison with the control group and showed that the T. catharinensis had antimutagenic activity within the type, time and form of treatment. Since the germination test showed that the conidia germination was accelerated from the bud phase, activities at the cell cycle level and polarized growth proved to be possible. The morphoanatomic analysis of the leaf and stem associated with the mutagenic and antimutagenic analyses contributes to the safe use of the plant by humans and also for the quality control of a possible phytotherapeutic drug.


RESUMO Tabernaemontana catharinensis A. DC (Apocynaceae) é utilizada como planta medicinal pela população. A fim de contribuir para o uso seguro da planta como medicinal, este trabalho teve como objetivo caracterizar morfoanatomicamente os órgãos vegetativos aéreos de T. catharinensis e avaliar a atividade mutagênica e antimutagênica de suas folhas. Foram realizados cortes histológicos da folha e do caule de T. catharinensis e, para a avaliação mutagênica e antimutagênica, foi utilizado o sistema metionina (methG1) e análise da germinação de conídios em Aspergillus nidulans. A análise morfoanatômica evidenciou a ausência de tricomas no caule, pecíolo e folha; presença de feixes bicolaterais, com exceção no ápice foliar cujos feixes são do tipo colateral; folha anfiestomática com estômatos paracíticos e camada subepidérmica na região do bordo foliar. O ensaio de mutagenicidade/antimutagenicidade mostrou uma diminuição significativa da frequência de mutação em relação ao controle, indicando que nesse tipo, tempo e forma de tratamento, T. catharinensis apresentou atividade antimutagênica. O ensaio de germinação evidenciou que houve aceleração da germinação dos conídios, a partir da fase de botão, indicando uma possível atuação em nível de ativação de ciclo celular e crescimento polarizado. A análise morfoanatômica da folha e do caule associados à análise mutagênica e antimutagênica, contribuem para o uso seguro da planta pela população e para o controle de qualidade de um possível fitoterápico.

Introdução: Bellis perennis L. (Asteraceae) é conhecida popularmente como margarida, margarita, margarida-vulgar, margarida-menor ou margarida-comum. É uma herbácea perene, amplamente distribuída em toda a Europa e Norte da África. Objetivo: realizar um levantamento bibliográfico sobre aspectos agronômicos, genéticos, morfoanatômicos, químicos, toxicológicos e farmacológicos desta espécie. Métodos: realizada uma revisão dos artigos completos originais e de revisão disponíveis na literatura, por meio de busca de artigos nos idiomas português, inglês ou espanhol, compreendidos entre os anos de 1962 e 2011 nas seguintes bases de dados: Literatura Latino-Americana e do Caribe em Ciências da Saúde, Science Direct, Scielo e Medical Literature Analysis and Retrieval Sistem on-line. Foram utilizados, para busca dos artigos, os seguintes descritores: Bellis perennis, B. perennis, Asteraceae, estudos botânicos, estudos químicos, estudos toxicológicos e atividades farmacológicas. Além disso, foram incluídos os resultados experimentais dos estudos de toxicidade aguda em camundongos tratados com extrato etanólico das flores de B. perennis. Resultados: sua ampla utilização como planta medicinal e devido à ausência de informações sistematizadas sobre sua toxicidade e ações farmacológicas justificam a importância dessa revisão, uma vez que já foram isolados vários constituintes químicos. Em virtude dessas propriedades terapêuticas ainda não serem bem compreendidas, e devido à sua importância na fitoterapia, novos estudos fitoquímicos e farmacológicos devem ser conduzidos. Conclusão: apesar do seu amplo uso, a literatura não fornece detalhes sobre as suas propriedades toxicológicas e farmacológicas, demonstrando a uma necessidade de novas pesquisas científicas.

Objetivos: realizar una revisión de la literatura sobre los aspectos agronómicos, genéticos, morfológicos y anatómicos, químicos, toxicológicos y farmacológicos de esta especie. Métodos: se llevó a cabo una revisión de los artículos completos originales, así como de la literatura disponible en portugués, inglés o español, entre 1962 y 2011, en las bases de datos siguientes: LILACS, Science Direct, Scielo y Medical Literature Analysis and Retrieval Sistem on-line. Se utilizó para buscar artículos, los descriptores siguientes: Bellis perennis, Bellis perennis, Asteraceae, estudios botánicos, químicos, estudios toxicológicos y actividades farmacológicas. Adicionalmente, se incluyeron los resultados de los estudios experimentales de toxicidad aguda en ratones, tratados con el extracto de etanol de flores de Bellis perennis. Resultados: su amplio uso como planta medicinal y por causa de la falta de información sistemática sobre su toxicidad y acciones farmacológicas, se justifica la importancia de esta opinión, porque ya se han aislado varios componentes químicos. Como estas propiedades terapéuticas aún no se comprenden bien, y por su importancia en fitoterapia, nuevos estudios fitoquímicos y farmacológicos deben realizarse. Conclusión: a pesar de su uso generalizado, la literatura no proporciona detalles acerca de sus propiedades toxicológicas y farmacológicas, lo que demuestra la necesidad de la investigación científica.


Introduction: Bellis perennis L. (Asteraceae), commonly known as daisy, is a perennial herb of wide distribution throughout Europe and the north of Africa. Objectives: conduct a bibliographic review about the agronomic, genetic, morphological and anatomic, chemical, toxicological and pharmacological characteristics of this species. Methods: a review was carried out of full-text original papers, as well as materials published in Portuguese, English and Spanish between 1962 and 2011 in the following databases: LILACS, Science Direct, Scielo and Medical Literature Analysis and Retrieval System on-line. The search terms used were Bellis perennis, B. perennis, Asteraceae, botanical studies, chemical studies, toxicological studies and pharmacological activities. We also included the results of experimental studies on acute toxicity in mice treated with ethanolic extract of flowers of Bellis perennis. Results: its wide use as a medicinal plant and the scarcity of systematic information about its toxicity and pharmacological actions, justify the relevance of this review, since several chemical components have already been isolated. Given the fact that these therapeutic properties are still not well understood, and in view of their importance for phytotherapy, it is recommended that further phytochemical and pharmacological studies be carried out. ABSTRACT

Euphorbia hirta L., popularmente conhecida por erva andorinha, tem sido utilizada no tratamento de distúrbios respiratórios e como antiespasmódico. Esse trabalho foi realizado com a finalidade de determinar as características macroscópicas e microscópicas das folhas, caule e raiz dessa planta, uma vez que estas são as partes usadas popularmente. Algumas características de valor na sua diagnose foram apontadas e ilustradas por fotomicrografias. Reações de fitoquímica e a determinação do conteúdo de água e de cinzas foram realizadas no pó obtido da planta inteira, parte essa utilizada na fitoterapia Ayuverda. Na prospecção fitoquímica foram encontrados flavonóides, mucilagem, resinas e cumarinas. Na determinação do teor de água, de cinzas totais e de cinzas insolúveis em ácido, os valores encontrados foram 8,99%, 9,35%, 0,75%, respectivamente. Com o desenvolvimento deste trabalho foi possível obter parâmetros para o controle de qualidade das folhas, caule e raízes de E. hirta.


The Euphorbia hirta L. has been used in the treatment of respiratory disorders and as an antispasmodic. This work was carried out to determine the macroscopic and microscopic characteristics of the leaves, stems and roots of this plant, as these parts are commonly used. Some features of value in their diagnosis were identified and illustrated by photomicrographs. Phytochemical reactions and determination of water and ash contents were carried out in the powder obtained from the whole plant, the part used in Ayurveda phytomedicine. In phytochemical screening, we found flavonoids, mucilage, resins and coumarins. The values of water content, total ashes and acid insoluble ashes were 8.99%, 9.35%, 0.75%, respectively. The data obtained are important for the quality control of the leaves, stems and roots of E. hirta.

ABSTRACT

Gochnatia polymorpha (Less.) Cabrera, Asteraceae, is popularly known as cambará and cambara-de-folha-grande in Brazil. It is used in traditional medicine to treat respiratory and gastrointestinal disorders. Pharmacological studies revealed anti-inflammatory, antispasmodic, antibacterial and antiviral activities. The goal of this paper was to carry out morphological and anatomical studies in order to describe the aerial parts of G. polymorpha. The botanical material was collected, fixed, and prepared according to usual light and scanning electron microtechniques. The leaves are simple, oblong-lanceolate to elliptical-lanceolate in form with mucronate acute apex, rounded base, entire or slightly toothed margin, and short petiole. In transection, the epidermis is uniseriate along the leaf blade. A subepidermal layer next to the adaxial side is present. Anomocytic stomata are seen only on the abaxial surface. Capitate glandular trichomes and T-shaped non-glandular trichomes occur on the leaves. The mesophyll is dorsiventral and minor collateral vascular bundles are enclosed by a sheath of thick walled parenchymatic cells. The midrib is biconvex and the petiole has a circular shape. The epidermis of the stem consists of a single layer of cells with glandular and nonglandular trichomes. The vascular cylinder shows typical structure and perivascular fiber caps are next to the phloem.

ABSTRACT

This study aimed to evaluate the existence of leaf anatomic characteristics in Tabebuia alba changed by air pollutants, which could be used as tool for a bioindication program. The quantification of mutagenic events on pollen grains also were measured. For this, median leaves and pre-anthesis flowers were collected from the adult plants from three places of Curitiba and one place in Araucaria, all nearby to the air monitoring stations. The comparison of the four study sites showed a reduction in leaf area, an increasing of stomatal density, subepidermic layer, epidermis in both faces and the amount of micronucleus. Also, there was reduction of chlorophyllian parenchymas at the site where there was the higher average for the ozone level. It was concluded that these modifications might be a consequence of the effect of troposferic pollution on T. alba plants. However, further studies with this species would be necessary to confirm its potential for bioindication.

Many species of Myrtaceae, including Plinia edulis (Vell.) Sobral (cambucá), have pharmacological properties and are used as hypoglycemiants and therapeutic agents against stomach problems and throat infections. Samples were collected from Tijuca Forest in Rio de Janeiro, and the morpho-anatomical data were compared with other specimens obtained from Trindade, Paraty, found in the literature. Variations in leaf anatomy were observed, and the possible causes for these effects are discussed. The plant material collected from Tijuca Forest was analyzed using scanning electron and optical microscopy. Histochemical tests were applied to identify starch, lipids, phenolic compounds and lignin. The epidermal cells exhibit straight or slightly sinuous anticlinal walls covered by a smooth cuticle with granules of wax. Simple trichomes are restricted to the midrib region, and paracytic stomata are only observed on the abaxial leaf surface. The mesophyll is dorsiventral, with conspicuous intercellular spaces in the spongy parenchyma. Intercalated columns of crystalliferous cells and subepidermal secretory cavities are observed in the single layer of palisade parenchyma. The samples obtained from Trindade, Paraty, show larger leaves, anomocytic stomata and trichomes scattered throughout the leaf surface. This plasticity might reflect leaf adaptations to environmental factors or different stages of leaf development.

ABSTRACT

Erythrina velutina Willd., Fabaceae, known as "mulungu", is a tree of tropical regions, as northeastern Brazil. Its bark is used in folk medicine as tranquilizer, sedative and insomnia. This study aimed to characterize the stem and leaf anatomy and to provide subsidies to quality control of the plant drug due to its wide use in folk medicine as well as its differentiation from other species with the same popular name. Samples were collected at Cuité, in Paraíba State, Brazil, fixed in FAA50, semipermanent slides were made, following usual procedures in plant anatomy. The stem shows a cylindrical contour, covered by a uniseriate epidermis covered by a thickened cuticle. It shows claviform glandular and branched trichomes with uniseriate stalk. Secretory cavities are into the phloem. The leaf epidermis has branched and glandular trichomes and anisocytic and paracytic stomata, on both sides, with predominance of branched trichomes and stomata on abaxial surface. Secretory cavities in stem and leaf, types of trichomes and stomata, its location and distribution constitute diagnostic characters for this specie. The structural characterization of the stem and leaf allows its distinction from other ones of this genus, ensuring safety for commercial pharmacological uses, allowing certification of the authenticity of raw material.

Croton echioides Baill., Euphorbiaceae, is a small tree found in Bahia, Northeastern Brazil. Its stem bark has been widely sold as an aphrodisiac and tonic, as a substitute for the roots of Ptychopetalum olacoides Benth. Olacaceae, the Amazon Muira Puama or Marapuama, and C. echioides is characterized as the "Northeastern Marapuama". This contribution describes a morphoanatomical analysis and pharmacognostic study of stem bark of this species. The stem has a thick cortex with compound starch grains and laticifers; a sclerenchymatic sheath which consists of brachysclereids with large crystals externally to the phloem, and abundant fiber in the secondary xylem, as the main features of the species. The data obtained for water content (9.26±0.07%), water-soluble extractives (3.92±0.19%), total ash (1.24±0.06%) and acid-insoluble ash (0.16±0.01%), together with the chromatographic profile obtained by TLC, contribute to the quality control and standardization for the plant drug. The pharmacological screening indicated LD50 values above 500 mg/kg orally and equal to 500 mg/kg by the i.p. route, as well as some stimulant potential, depending on the dose.

Centrolobium tomentosum Guillemin ex Benth., Fabaceae, known as "araribá-rosa", is a large tree that is widespread throughout Brazil. The bark of its stem is used in folk medicine as an astringent and the leaves are employed as a poultice for wounds and bruises. The aim of this study was to contribute to the pharmacognostic quality control of this medicinal species through a macro- and microscopic diagnosis of its leaves and stems. Mature leaves and young stems were collected at Embrapa (Colombo, PR) and analyzed using standard microtechniques. The leaves are opposite or alternate, compound, imparipinnate, oblong-lanceolate and hypostomatic with paracytic stomata. The mesophyll is dorsiventral and the midrib is biconvex with a collateral vascular bundle in a circular arrangement. The petiolule is circular and its vascular system is composed of two major vascular bundles in the middle, with smaller bundles between them. The rachis has a structure similar to the petiole, and these parts have a sclerenchymatic sheath shaped in multiple arcs, which surrounds a collateral vascular bundle that is centrally located. The stem has a uniseriate epidermis with peripheral phellogen and a sclerenchymatic sheath composed of many arcs, which surrounds the phloem. The stems and leaves also have non-glandular and glandular trichomes, phenolic compounds and idioblasts made of calcium oxalate crystals. This manuscript has an important role in the morpho-anatomical diagnosis of the Brazilian flora.

Objectives: To study the morpho-anatomy of the aerial parts of Rivea hypocrateriformi (Convolvulaceae) to increase the knowledge and standardization parameters of these plants and its family. Methods: Morpho-anatomical studies of aerial parts of have been carried out by free hand. The different types of histochemical tests were performed for the identification of micro-chemical by using staining reagents. Preliminary phytochemical and quantitative estimation has been determined along with HPTLC fingerprinting. CAMAG HPTLC system equipped with Linomat V applicator, TLC scanner 3, Reprostar 3 and WIN CATS-4 software was used for fingerprinting. Results: The morpho-anatomical structure of plant revealed the presence of thick wall collenchyma cell and parenchyma cells along with spharaphides and columnar palisade cells. Presense of numerous covering trichomes, parasitic stomata, oval shaped vascular bundles and calcium oxalate crystals were observed. In powder microscopy lignified xylem vessels, and covering trichomes were clearly seen. Physicochemical parameters, ash value, inorganic elements, moisture content and extractive value were determined to develop the stringent pharmacognostic standards. Qualitative standardization and HPTLC fingerprinting of phytochemical revealed the presense of various plant secondary metabolites. Conclusions:These findings will be useful in establishing pharmacognostic and phytochemical standards for identification, as well as assessment of purity and quality of this plant, which definitely gaining the relevance in plant drug research and establishment of plant monograph.
